General Information

Mice: C57/BL6

Expression the human wild type PARKIN gene unilaterally in the SN using recombinant Adenovirus-Associated virus 2/8 (AAV 2/8).

Endogenous Parkin: Yes

Corresponding human genotype: Parkin was the first gene associated with autosomal recessive Parkinson’s disease (PD).

Targeted gene: Parkin

Viral injection

Injection coordinates: right SN -. one deposition site: AP −3.1 mm, ML −1.2 mm, DV −4.0 mm.

Volume of injection and viral titer: Two microliters of rAAV vector is injected at a concentration of 2 × 1011 GC/ml

Transgene expression

The level of transgene expression is unknown, however, transduction efficacy of the nigral dopaminergic cells reaches 90%.

Neurodegeneration

12 months: No visible degeneration can be observed between the mice injected with the recombinant viral vectors containing the human Parkin or GFP.
